If my opponent uses an effect that states "when this ninja is put in play, randomly discard 1 card from your opponents hand."

they must declare a random card in my hand as a target correct?

so if they choose a card, and i decide to chain a mission by using that chosen card as a hand cost, since their target is no longer in my hand, does their effect miss and fail?

No you don't choose a target unless a card asks for a target. Your example does not ask for a target, there for no target is selected.
